mRNA vaccines introduce mRNA encoding antigenic proteins into the body, which undergo direct translation to form corresponding antigenic proteins, thereby inducing a specific immune response in the body to achieve the effect of preventive immunity.

Upon entry of exogenous mRNA into the body, it activates the immune system, triggering inflammatory reactions caused by the mRNA itself, and may lead to clearance of the mRNA before it can exert its effect. Therefore, introducing naturally modified nucleotides is one of the effective means to reduce the immunogenicity of mRNA vaccines.

Pseudouridine is the most abundant modified nucleotide on RNA, also known as the "fifth nucleotide" of RNA. In 2005, Katalin Karikó and others discovered that introducing pseudouridine into RNA can reduce its immunogenicity, and the immunogenicity of RNA decreases with increasing proportions of pseudouridine. In 2008, Katalin Karikó and others found that using pseudouridine to completely replace uridine in mRNA not only greatly reduces the immunogenicity of mRNA but also enhances its stability and translation ability. In 2015, Oliwia Andries and others found that using N1-methyl pseudouridine to completely replace uridine can further reduce the immunogenicity of mRNA and enhance its protein expression ability compared to using pseudouridine alone.
